Abstract:
Electron microscopic studies of tissue fixed in formalin and stored for a long time, enabled some peculiar forms of pathologic tissue, such as extremely rare neoplasms, to be investigated. Tissues were washed for 3 to 4 hr in a 4.5% sugar solution prepared in a phosphate buffer pH 7.4; then they were fixed for 12 to 24 hr in an 0.5% solution of OsO4 prepared in this sugar solution with the addition of K2Cr2O7 up to a concentration of 0.5%. In the course of the treatment the sections were contrasted in a piece of uranil acetate added to the third portion of 70° alcohol up to a concentration of 1%; epon or araldite were poured over and the sections were once more contrasted on a screen for a maximum of 10 min with a warm solution (25°C) of lead citrate.
